Courses@CRG: Biological Super-Resolution Microscopy

In the last years, super-resolution light microscopy equipment has become more widely available for biological researchers. Most if not all of the existing approaches do however present challenges in sample preparation, image acquisition and also the subsequent analysis that need to be met for the successful execution of a biological experiment. The course will provide an introduction into super-resolution imaging for STED and localization microscopy on state of the art instrumentation and focus on the important aspects of label selection, labeling strategies, sample mounting, imaging (3D) and subsequent anaylsis of STED and dSTORM datasets.

  • Labeling considerations, imaging buffers and mounting media for super-resolution microscopy: localization microscopy and STED
  • Stimulated emission depletion (STED) microscopy
    • Pulsed, CW and time-gated STED imaging
    • Multichannel STED imaging
    • 3D STED imaging
    • in vivo STED imaging
  • Localization based super-resolution microscopy methods
    • Multichannel 3D direct Stochastic Optical Reconstruction Microscopy (dSTORM)
    • Point Accumulation for Imaging in Nanoscale Topography (PAINT)
  • Rendering and processing of super-resolution datasets
  • Deconvolution of 3D STED datasets
  • Rendering of point-based SR data
  • Registration of image channels
  • Adaptive Optics in SR microscopy
